You can tell the windows mixer to send the Spotify audio to one of the Voicemeeter inputs (with a virtual cable, more info on their site) and then configure a macro to control that input volume with the knob. On voicemeeter, there are two knobs above the volume meter labeled comp & gate. Adjust the gate knob to remove noise. It is a noise gate. Basically, you want to raise that as high as possible without it reducing the sound you want or cutting out the sound you want.
